β3-Adrenoceptor detection and signal transduction: focus on antibody validation and urinary bladder

β3-Adrenoceptors are believed to play a role in the functional regulation of a variety of organs and tissues including the brain, heart, blood vessels, uterus and urinary bladder. Recently, it has been recognized that activation of these receptors can be beneficial in the treatment of patients suffering from an overactive bladder (OAB), and one compound, activating β3-adrenoceptors, mirabegron, is now available in many countries as a medicine to treat OAB. However, we still have a limited understanding of the occurrence and the density of β3-adrenoceptors in the human body.
The research described in the thesis of Hana Černecká focused on the elucidation of signaling mechanisms following β3-adrenoceptor activation of the urinary bladder, as well as the detection of β3-adrenoceptor proteins in various tissues .
OAB is highly prevalent and reduces health-related quality of life. Currently, it is mainly treated with medicines inhibiting muscarinic receptors, but their use is limited due to side effects like dry mouth, blurred vision and constipation. Thus, stimulation of β3-adrenoceptors provides an alternative, noveloption for treating symptoms of OAB.
Černecká and her co-authors provided evidence about the molecular mechanisms involved in the relaxation induced by activation of β3-adrenoceptors in the urinary bladder of rats and humans. Importantly, β3-adrenoceptor activation was found to have stronger effects against stimuli occurring in diseased than in healthy bladder. Moreover, specific antibodies raised against β3-adrenoceptors were identified and validated for detection in human and rodent tissues.
