High yield harvesting of bacterial outer membrane vesicles for the treatment of meningitis

    Abstract

    In this thesis, we developed a magnetic harvesting method for bacterial OMVs, which significantly enhances their yield and endows OMVs with magnetic responsiveness. We demonstrated that magnetic OMVs can be guided via magnetic fields for targeted drug delivery both in vitro and in vivo. Antibiotic and sonosensitizer-loaded magnetic OMVs achieved excellent drug delivery in the brain of mice for the treatment of meningitis with better therapeutic efficacy and reduced recurrence of meningitis by more effectively killing antibiotic-tolerant bacteria than free drugs alone.
