    Samenvatting

    In mass electrofusion systems with aggregation of protoplasts by alignment, the yield and composition of fusion products can be predicted by a simple model. Through computer simulation, upper limits were found for the yield of binary and multi fusions. To overcome constraints on binary products, surface labelling of plant protoplasts with biotin and avidin was applied to control the aggregation process. Avidin-labelled protoplasts associated with biotinylated protoplasts when combined. Within aggregates of a heterologous combination, more heterologous than homologous contacts between protoplasts occurred. Electrofusion could be induced in the aggregates and fusion events were shown to depend on the angle in the electric field. Also, PEG fusion could be induced. Due to the shift towards heterologous contacts, an increase in heterofusion yields with both fusion systems was observed. Likely applications of the aggregation protocol in various selection procedures for somatic hybrids (including bulk systems, now sorting and particle binding) are discussed.
