Samenvatting

The exceptional genomic content and genome organi-zation of the Acidianus filamentous virus 1 (AFV1) thatinfects the hyperthermophilic archaeon Acidianushospitalis suggest that this virus might exploit anunusual mechanism of genome replication. An analy-sis of replicative intermediates of the viral genome bytwo-dimensional (2D) agarose gel electrophoresisrevealed that viral genome replication starts by theformation of a D-loop and proceeds via strand dis-placement replication. Characterization of replicativeintermediates using dark-field electron microscopy, incombination with the 2D agarose gel electrophoresisdata, suggests that recombination plays a key role inthe termination of AFV1 genome replication throughthe formation of terminal loops.
